Rapid isolation of metaphase chromosomes containing high molecular weight DNA
Metaphase chromosomes with high molecular weight DNA were isolated from Chinese hamster ovary (CHO) cells in a neutral buffer containing polyamines and chelators. The individual, unfixed chromosomes retained their centromeric and secondary constrictions, distinct sister chromatids, and complex banding patterns. The DNA from these chromosomes was 100-fold larger (2 x 10(8) daltons) than DNA from...
متن کاملRapid isolation of high molecular weight DNA from marine macroalgae
Application of molecular techniques to study marine rnacroalgae is in its infancy, and is likely to be facilitated by the ability to routinely isolate high quality DNA from these plants. The generally high polysaccharide and polyphenol content in rnacroalgae, however, often interferes with the isolation and subsequent enzymatic manipulation of their nucleic acids. We describe the use of a CTAB ...
متن کاملSynthesis of high molecular weight polyethylene using FI catalyst
A FI Zr-based catalyst of bis[N-(3,5-dicumylsalicylidene)-2′,6′diisopropylanilinato]zirconium(IV) dichloride was prepared and used for polymerization of ethylene. The effects of reaction conditions on the polymerization were examined in detail. The increase in ethylene pressure and rise in polymerization temperature up to 35 oC were favorable for catalyst/MAO to raise the catalytic activity as...
متن کاملIsolation of intact high-molecular-weight DNA by using guanidine isothiocyanate.
A method for obtaining high-molecular-weight chromosomal DNA from Bacteroides intermedius and Bacteroides gingivalis is described. This technique is a modification of the guanidine isothiocyanate isolation procedure for RNA and should be useful for isolating intact DNA from organisms with high endogenous nuclease activity.
متن کاملDNA-templated synthesis in organic solvents.
DNA-templated organic synthesis (DTS) enables new modes of controlling chemical reactivity and allows evolutionary principles to be applied to the discovery of synthetic small molecules, synthetic polymers, and new chemical reactions. The structures that can be accessed in a DNA-templated format thus far have been limited to those that can be generated in aqueous solvents. Here we report effici...
